　　細胞凍存和復蘇實驗原理:細胞凍存及復蘇的基本原則是慢凍快融，實驗證明這樣可以大限度的保存細胞活力。目前細胞凍存多采用甘油或二甲基亞砜作保護劑，這兩種物質能提高細胞膜對水的通透性，加上緩慢冷凍可使細胞內的水分滲出細胞外，減少細胞內冰晶的形成，從而減少由于冰晶形成造成的細胞損傷。復蘇細胞應采用快速融化的方法，這樣可以保證細胞外結晶在很短的時間內即融化，避免由于緩慢融化使水分滲入細胞內形成胞內再結晶對細胞造成損傷。

　　細胞凍存和復蘇實驗注意事項:

　　1. 從增殖期到形成致密的單層細胞以前的培養細胞都可以用于凍存，但好為對數生長期細胞。在凍存前**好換一次培養液。

　　2. 將凍存管放入液氮容器或從中取出時，要做好防護工作，以免**。

　　3. 凍存和復蘇好用新配制的培養液。

　　當細胞冷到零度以下，可以產生以下變化：細胞器脫水，細胞中可溶性物質濃度升高，并在細胞內形成冰晶。

　　如果緩慢冷凍，可使細胞逐步脫水，細胞內不致產生大的冰晶;相反，結晶就大，大結晶會造成細胞膜、綱胞器的損傷和破裂。復蘇過程應快融，目的是防止小冰晶形成大冰晶，即冰晶的重結晶。
